Glycohyodeoxycholic Acid (GHDCA) is a metabolite in humans and a glycine derivative of the secondary bile acid (BA) hyodeoxycholic acid (HDCA).
In liver hepatocytes, hyodeoxycholic acid and other secondary bile acids (BAs), undergo a process of conjugation with amino acids glycine or taurine, catalyzed by specific enzymes.
Glycohyodeoxycholic Acid’s precursor compound - the secondary BA hyodeoxycholic acid, is a conjugate of hyodeoxycholate and derives from cholic acid. Hyodeoxycholic acid belongs to the group of 5ꞵ-cholanic acids, 6⍺,20-murideoxycholic acid, and C24-steroids. Apart from being present in humans, it’s also found as a metabolite in mice. HDCA is known to increase in patients with ulcerative colitis or Crohn’s disease. It’s also been studied that in LDL receptor knockout mice on the Western diet, the secondary BA reduces levels of LDL and VLDL, alongside fasting glucose concentrations and atherosclerotic lesions. HDCA is a TGR5 (GPCR19) agonist, as well. This is a G-protein BA receptor, involved in numerous signaling pathways, while it also acts as a metabolic regulator in cholesterol, glucose and energy, and BA metabolism and homeostasis. The receptor is associated with inflammatory response, liver regeneration, and cancer growth.
Thus, HDCA and its derivatives, including Glycohyodeoxycholic Acid, play an important role in the normal occurrence of a range of metabolic processes in the organism.
In addition, studies show that Glycohyodeoxycholic Acid administration in lithogenic diet-fed prairie dogs prevents the formation of cholesterol crystals in the gallbladder and decreases the activity of cholesterol 7α-hydroxylase.
