Procedure for TLC of synthetic PIPs
- Prepare 5% aqueous solution of potassium oxalate
- Dip TLC plate in potassium oxalate solution
- Dry and activate the TLC plate
- Lightly spot the plate with lipid; do not apply too much lipid to plate as it will cause streaking.
- Develop plate in chloroform:methanol:water:acetic acid (10:10:3:1, v/v)
- Visualize spots with suitable detection system, e.g., iodine vapor or molybdate spray
- Rf of PIPs (these values are reference points; actual results may vary depending on activity of plate and environmental conditions):
- PIP2: 0.3-0.4
- PIP3: ~0.2
