MRI Imaging Reagents

Gadolinium (Gd)-based contrast agents are used frequently in magnetic resonance imaging (MRI). Liposomes are often studied as potential carriers of MRI contrasting agents and drug molecules. Liposomes represent an exciting area of study regarding the generation of targeted diagnostic systems. There are several advantages to using Gd-chelating lipids as nanocarriers. Liposomes composed of Gd-chelating lipids, such as PE-DPTA (Gd), are easily modified to capture targets on their surface. The surface of these liposomes can be coated with a number of molecules to achieve targeting and increased circulation properties which are necessary for carriers of contrast agents in MRI. Recently, the safety of an MRI imaging liposomal platform containing Gd+3 chelator lipid PE-DTPA, DSPC, and cholesterol were tested in vitro for potential toxicological effects. The study found no adverse impact on human-derived hepatocyte-like HepaRG cells and macrophages indicating that further preclinical testing on animal models is warranted.
