Lipid 5 is a synthetic ionizable lipid with a pKa value around 6.5 that is predominantly used in the formulation of LNPs to deliver messenger RNA (mRNA). Lipid 5 LNPs result in the expression of multiple proteins while also clearing from tissue quickly. Lipid 5 has been shown to accumulate predominantly in the liver and spleen with lesser amounts detected in the lungs and kidneys. Another key feature of Lipid 5 is its endosomal escape activity compared to other Ionizable lipids which is a measurement of how well a lipid can deliver cargo to the cytosol within a cell.1,2
