RBM1-151 is a coumarinic 1-deoxydihydroceramide that can be hydrolyzed by acid ceramidase, allowing for fluorescence measurement upon cleavage and thus enabling the quantification of enzyme activity. Due to this fluorescent ability, RBM1-151 is utilized in research to monitor the activity of lipid amidases within live cells. By combining RBM1-151 with specific irreversible inhibitors of AC (such as SACLAC) and FAAH (like URB597), researchers can estimate the activities of AC, NAAA, and FAAH in a single experiment. This capability is particularly useful in studies involving cancer cell lines, such as acute myeloid leukemia (AML) and melanoma, where these enzymes play significant roles.
