n-dodecyl-ß-D-maltoside (DDM), or n-dodecyl-β-D-maltopyranoside, is a high-purity, non-ionic detergent extensively used to study the solubilization, stabilization, and structure of membrane proteins. Unlike harsh detergents that can denature proteins, DDM gently disrupts lipid bilayers while preserving protein functionality. This has made it an essential tool in biochemistry, structural biology, and pharmaceutical research.

With a low critical micelle concentration (CMC) of approximately 0.17 mM and a high aggregation number, DDM forms exceptionally stable micelles that enhance protein solubility and long-term stability. Its mild amphiphilic nature makes it a preferred detergent in cryo-EM, X-ray crystallography, and NMR studies—where maintaining native protein confirmation is highly important.

Application

DDM is widely used for isolating and purifying membrane proteins—including G-protein coupled receptors (GPCRs) and ion channels—to ensure structural integrity and functional activity. It’s frequently used in X-ray crystallography, nuclear magnetic resonance (NMR), and electron microscopy to facilitate high-resolution structural analysis.

Another notable characteristic of n-dodecyl-β-D-maltopyranoside is its ability to support the formation of proteoliposomes and model membrane systems to aid in studying lipid-protein interactions. Additionally, it improves the solubility and activity of enzymes such as lipases, ATPases, and cytochrome complexes, improving assay reliability and reproducibility.
