Tauroisolithocholanoic acid
5β-cholanic acid-3β-ol-N-(2-sulphoethyl)-amide, sodium salt
Tauroisolithocholanoic Acid is an isomer of the naturally occurring taurolithocholic acid (taurolithocholanoic acid) and has a role of a human metabolite.
Taurolithocholic acid is a bile salt that is a taurine-derivative of lithocholic acid. Lithocholic acid itself is a secondary and an end product of the bacteria metabolism in the gut, with a range of biological properties.
Bile acids are steroid amphipathic molecules, synthesized exclusively in the liver hepatocytes from cholesterol metabolism in a series of enzymatic reactions. They serve as potent digestive detergents that facilitate the absorption and transport of lipids, sterols, and fat-soluble vitamins in the intestines and liver.
Over the past decade, it has been found that BAs can also be considered hormones that regulate a range of metabolic pathways. Not only do they control their own homeostasis and enterohepatic circulation, but they modulate triglyceride, cholesterol, glucose, and energy metabolism, as well. However, when BAs levels increase to abnormally high concentrations, they become cytotoxic, inducing oxidative stress, mitochondrial and DNA damage, disruption of metabolic processes, and causing eventual liver fibrosis and cirrhosis.
As a bile salt derivative, Taurolthocholic acid owns similar properties in the human body in addition to some specific roles and functions. The bile salt can also solubilize fats and act as a cholagogue and choleretic.
On the other hand, in experimental research, tt is often used to induce and investigate cholestasis in ex vivo and in vivo animal models.
As an isomer, Tauroisolithocholanoic Acid can serve as an important detergent to explore the structure and functions of the tauro-conjugate taurolithocholanoic acid. Isomerism is crucial in pre-clinical medicine and pharmacology to determine the pharmacokinetic and pharmacodynamics of organic and synthetic compounds.
