Cholic acid 7-sulfate, also known as 7-sulfocholic acid or 7-sulphocholat, is a metabolite of one of the primary bile acids - cholic acid, derived by conjugation to neutralize accumulated toxic bile acids.
Bile acids (BAs) are biomolecules, synthesized in the liver as a result of cholesterol oxidation, and are secreted in the duodenum. BAs own a range of important physiological properties including regulation of gene expression, involved in lipid, glucose, energy, and their own homeostasis. However, their abnormally high concentration in some specific conditions can have pathological consequences, like liver cytotoxicity or carcinogenicity. Therefore, BA homeostasis is crucial to maintaining their physiological functions and preventing their pathologic effects.
The metabolic process sulfation is one of the major mechanisms to neutralize toxic BAs. Products of the reaction, catalyzed by the enzymes sulfotransferases (SULTs), are derivatives such as Cholic acid 7-sulfate. These sulfated BAs (SBAs) are less toxic than unsulfated ones because they have better water solubility, thus are easily excreted in urine and feces under pathologic conditions, like intrahepatic cholestasis. Hence, sulfation alongside its sulfate products, including Cholic acid 7-sulfate is an important BAs detoxification, elimination, and homeostasis metabolic pathway.
Moreover, unlike nonsulfated BAs, Cholic acid 7-sulfate and other sulfated derivatives are resistant to hydrolysis and metabolism by the intestinal microflora, which inhibits their intestinal reabsorption providing protection of the enteric mucosa.
