Taurolithocholic Acid (TLCA), or Lithocholyltaurine, Taurolithocholate, and 5ß-cholanic acid-3α-ol N-(2-sulphoethyl)-amide, sodium salt, is a bile acid taurine conjugate and a key human metabolite involved in lipid digestion and absorption. As a component of bile, TLCA assists in fat and sterol emulsification and helps facilitate their absorption in the intestines.
Beyond its role in digestion, TLCA has been widely studied for its physiological and pharmacological effects. Research indicates that it can modulate apoptosis by increasing caspase-3 and caspase-7 in hepatitis B virus (HBV)-infected hep3B cells. Additionally, it serves as an agonist of TGR5, a bile acid receptor belonging to the GPCR family that promotes energy expenditure in brown adipose tissue.
Application
Taurolithocholic Acid is a powerful cholestatic agent that can hinder bile flow, disrupt hepatobiliary exocytosis, and alter the insertion of transport proteins in hepatocyte membranes. Emerging studies suggest that it may have therapeutic applications in severe fever with thrombocytopenia syndrome virus (SFTSV) infection, where it has demonstrated antiviral and anti-inflammatory properties.
Furthermore, TLCA is frequently used in experimental models to induce cholestasis and pancreatitis, making it useful for liver and pancreatic disorder studies.
