Tauroursodeoxycholic Acid (TUDCA), also known as Ursodeoxycholyltaurine, is the taurine-conjugated and more hydrophilic form of the secondary bile acid (BA) ursodeoxycholic acid (ursodiol). TUDCA is an amphiphilic BA, present in trace amounts in the body. The metabolite has been found to show anti-inflammatory, neuroprotective, and cardioprotective abilities, and acts as a powerful apoptosis inhibitor and a bone density conservation agent. As for its primary source, ursodiol, it’s a naturally occurring BA in human organisms and is widely used in cholestasis (failed bile excretion and BA back up in the liver) treatment and gallstone dissolution.
As an ursodiol derivative, Tauroursodeoxycholic Acid also shows great potency in treating cholestasis, since it has a higher water solubility, and soluble bile salts and conjugates counteract the toxicity of regular BAs accumulated in the liver. The taurine derivative is thought to protect the liver and help hepatocytes to regenerate.
Additionally, TUDCA seems to reduce stress on the endoplasmic reticulum organelle in all cells. This creates a range of beneficial conditions for important metabolic pathways. So, TUDCA eventually decreases insulin resistance and the risk of diabetes.
Furthermore, due to its neuroprotective properties, TUDCA is being investigated for use in the treatment of Alzheimer's disease. TUDCA administration in mouse models enhances Akt/GSK3β signaling pathway, which results in attenuation of tau pathology, glial activation, loss of synaptic function, and improved spatial memory. Tauroursodeoxycholic Acid can also directly protect mitochondrial membranes, thus aiding mitochondrial health, which is another mechanism of neuroprotection.
In research work on mouse models of Huntington’s disease, TUDCA has been again reported to manifest neuroprotective effects.
Tauroursodeoxycholic Acid is a very promising molecule with a lot more potential yet to be discovered.
