Glycocholic Acid-d4 is the deuterated form of the metabolite glycocholic acid. This means a hydrogen atom in the molecule of glycocholic acid has been replaced by a deuterium atom - deuterium is one of the two stable and heavy hydrogen isotopes.
Glycocholic acid, also known as cholylglycine, is a crystalline bile acid (BA), found as a sodium salt in the bile of mammals. It’s involved in the emulsification of fats. The compound is a secondary BA and its anions are called glycocholates.
Glycocholic acid is derived from the BA cholic acid and glycine - a nonessential amino acid, though a process of conjugation, carried out by specific enzymes from the colonic bacterial flora.
Glycocholic acid precursor - cholic acid, is one of the two primary bile acids in the human organism, along with chenodeoxycholic acid. Both BAs are obtained from cholesterol in pericentral liver hepatocytes of the hepatic acini. They occur in equal concentrations in the bile. Derivatives of cholic acid, including glycocholic acid, are derived from cholyl-CoA, which exchanges CoA with an amino acid.
As a glycine conjugate of cholic acid, glycocholic acid acts as a detergent to solubilize fats, fat-soluble vitamins, and other sterols for absorption.
The metabolite is often used as a biological detergent, as well.
It’s also studied to reduce the expression of the gene encoding the farnesoid X receptor (FXR) while it increases the expression of genes that encode bile acid receptors TGR5 and S1PR2 in SNU-245 cells when administered at a concentration of 1.6 μmol/ml (insert the references for these statements since they are very technical)
Glycocholic acid is found to stimulate the accumulation and cytotoxicity of Epirubicin - an anthracycline drug used in chemotherapy.
