Ionizable lipids are designed to be pH-sensitive, which is essential for effective delivery. They stay neutral at physiological pH (around 7.4), helping your LNP formulation stay stable in circulation. But once the LNP is taken up by a cell and enters the more acidic endosome, these lipids become positively charged, which helps the particle escape into the cytosol and release its nucleic acid cargo. That pH-triggered charge shift is critical — it’s what makes efficient delivery possible.
One of the biggest factors to consider when choosing an ionizable lipid is its apparent pKa — basically, how it behaves in the context of the full LNP formulation. Most ionizable lipids used in FDA-approved therapeutics have apparent pKa values between 6.0 and 7.0. That range helps balance stability in the bloodstream with the ability to release cargo inside cells.
